dc.description.abstract | Proteinasen verschiedener Proteinasefamilien zeigen eine negative Korrelation von Proteinaseaktivität und Abnahme der Schaumstabilität in Bier. Ein eindeutig negativer Einfluß auf die Schaumstabilität wurde sowohl für Proteinasen der Carboxylproteinasefamilie (Proteinase A und Pepsin), als auch der Serinproteinasefamilie (Carboxypeptidase Y) nachgewiesen. Die schaumnegativen Konzentrationen der Hefeproteinasen Proteinase A (PrA) und Carboxypeptidase Y (CPY) waren so gering, daß ein Nachweis mit herkömmlichen Methoden zur Konzentrationsbestimmung nicht möglich war. Von den Proteinhauptkomponenten im Bier unterliegt das schaumpositive Lipidtransferprotein 1 (LTP1) im Gegensatz zum Protein Z einem Abbau durch PrA. Damit wurde LTP1 eindeutig als spezifisches Substrat für PrA identifiziert. Verschiedene LTP1-Formen besitzen gegenüber PrA unterschiedliche Sensitivitäten. Während PrA ein in E. coli exprimiertes rekombinantes LTP1 abbaute, wurde der Abbau eines rekombinanten Histag-LTP1-Fusionsproteins und des nativen Gersten-LTP1 vermutlich konformationsbedingt behindert. Die proteinasesensitive LTP1-Form in Bier entsteht vermutlich durch Modifikationen während des Mälz- und Brauprozesses. Als Grundlage für die Entwicklung eines sensitiven Nachweises für PrA-Konzentrationen in Bier wurden potentielle PrA-Spaltsequenzen im LTP1-Molekül identifiziert. Massenspektrometrische Untersuchungen potentieller LTP1-Abbauprodukte ergaben, daß PrA in LTP1 sowohl Peptidbindungen C-terminal hydrophober (Ala, Ile, Leu) als auch polarer Aminosäuren (Gln, Tyr, Asp, Cys) hydrolysiert. | de |
dc.description.abstract | Proteinases of different families show a negative correlation between their activity and the reduction of foam stability in beer. For proteinases of the carboxyl proteinase family (e.g. Proteinase A and pepsin) and of the serine proteinase family a negative influence on foam stability has been shown. Conventional quantification methods were not sensitive enough to detect the low concentrations of Proteinase A (PrA) and Carboxypeptidase Y (CPY) that still exerted a foam negative effect. A major protein component in beer, the foam positive lipid transfer protein 1 (LTP1), has been shown to be degraded by PrA whereas another major protein component, the protein Z was not. This clearly identifies LTP1 as a specific substrate for PrA. Different forms of LTP1 possess distinct sensitivities with regard to PrA. While a recombinantly expressed LTP1 was degraded by PrA, the degradation of both a recombinant histag-LTP1 fusion protein and a native barley-LTP1 was presumably inhibited by their conformation. The proteinase sensitive form of LTP1 in beer is probably a result of modifications that occur during the malting and brewing process. Mass-spectroscopic analyses of potential degradation products of LTP1 showed that PrA hydrolyses peptide bonds C-terminal of hydrophobic (Ala, Ile, Leu) as well as of polar amino acids (Gln, Tyr, Asp, Cys). Potential cleavage sites for PrA in the LTP1-molecule were identified as a starting point for the development of a more sensitive quantification method for PrA in beer. | en |
