Selbstorganisation amphiphiler Block-Copolymere in Mikromischern

Die Selbstorganisation zweier Systeme amphiphiler Polymere, dem Diblock-Copolymer PB-PEO und dem Triblock-Copolymer PEO-PPO-PEO, wurde in Mikromischern erfolgreich untersucht. Es wurden drei Mikromischer eingesetzt, zwei auf dem interdigitalen Mischprinzip und einer auf dem Split-and-recombine-Prinzip basierend. Die jeweilige Mischerarchitektur wurde auf die Fähigkeit hin untersucht, Polymersomen kontrolliert und kontinuierlich herstellen zu können. Durch Veränderung der Mischbedingungen wie Mischerarchitektur, Flussraten, Flussratenverhältnisse, Konzentration, Temperatur oder Lösungsmittel-beschaffenheit, wurde die Selbstorganisation von PB-PEO gezielt gesteuert. Unterschiedliche Strukturen wie Polymersomen, scheibenartige Mizellen oder sphärische Mizellen wurden mit enger Größenverteilung hergestellt und mittels DLS und (Cryo)-TEM charakterisiert. Daraus ergab sich ein Einblick in den Mechanismus der Selbstorganisation zu Vesikeln. Gleichzeitige Einlagerung hydrophober Partikel wie Quantum Dots (QDs) oder Eisenoxid-Nanopartikel (USPIO-NPs) diente als Modellsystem für kontinuierlich hergestellte multi-funktionelle Polymersomen. Die Darstellung der PB-PEO-Vesikel wurde auf das Pluronic® System L121 erfolgreich übertragen. Eine durch hydrophobe Beladung entstandene Stabilisierung der sonst sehr instabilen Pluronic®-L121 Vesikel wurde erfolgreich nachgewiesen und lieferte kontinuierlich hergestellte biokompatible, multifunktionelle Polymersomen mit niedriger Polydispersität.
The self-assembly of two different amphiphilic polymer systems, the diblock- PB-PEO and the triblock-copolymer PEO-PPO-PEO, were investigated in micro mixers. Three different micro mixers were used: two based on interdigital mixing and one on split-and-recombine principle. The micro mixers were investigated to ensure a continuous and controlled synthesis of polymersomes. The self-assembly of PB-PEO was controlled by varying the mixing conditions like mixing architecture, flow rate, flow rate ratio, concentration, temperature or solvent properties. Different morphologies like polymersomes, disc-like micelles or spherical micelles were synthesized with a low polydispersity and characterized by DLS and (Cryo)-TEM. This gave an insight into the mechanism of polymersomes’ self-assembly. Simultaneous loading of hydrophobic particles like quantum dots (QDs) or iron oxide nanoparticles (USPIO-NPs) was used as a model system for continuously synthesized multifunctional polymersomes. Based on the mixing of PB-PEO, the controlled self-assembly was transferred to the pluronic® L121 system. A stabilization of the instable pluronic®-vesicles by loading of hydrophobic particles was successfully demonstrated and leads to a controlled and continuous synthesis of multifunctional, biocompatible polymersomes with a low polydispersity.
