Chemometrische Analysestrategien für Raman-spektroskopische Daten

Gegenstand dieser Arbeit sind chemometrische Methoden zur Analyse komplexer Datensätze, die mittels Raman-Spektroskopie erzeugt werden. Anhand exemplarischer Problemstellungen werden geeignete Strategien zur Dechiffrierung der chemischen und physikalischen Informationen eines biologischen oder chemischen Systems aus experimentellen Spektren dargestellt und bewertet. Gemeinsames Ziel der verwendeten methodischen Ansätze ist die Bestimmung der Eigenspektren der zum System gehörigen Spezies (Komponentenanalyse). Die so gewonnenen Komponentenspektren dienen dann der Bestimmung der quantitativen Verteilung zwischen den Spezies bzw. struktureller, thermodynamischer oder kinetischer Zusammenhänge. Die Arbeit zeigt die Vielfalt der Einsatzmöglichkeiten der quantitativen Spektrenanalyse und ihrer computergestützten Automatisierung. Am Beispiel der Qualitätskontrolle von Kaffeebohnen wurde gezeigt, dass zur qualitativen und quantitativen Unterscheidung zweier Spezies schon ein singulärer spektraler Marker ausreichend sein kann, dessen Natur nicht notwendigerweise bekannt sein muss. Durch den Einsatz algorithmischer Verfahren kann die Kontrolle statistisch aussagekräftiger Stichproben beschleunigt werden. Die Raman-Spektroskopie ermöglicht jedoch nicht nur die Unterscheidung von Spezies sondern auch ihre strukturelle Charakterisierung. Am Beispiel des biologischen Photorezeptors Rhodopsin werden Möglichkeiten der Strukturbestimmung bei strukturell variablen Systemen aufgezeigt und diskutiert. Die Ermittlung der Komponentenspektren von Spezies, die in einem Gleichgewicht vorliegen, eröffnet die Bestimmung thermodynamischer Zustandsgrößen. Als Beispiel wurden dazu die Raman-Spektren eines prototypischen Phytochroms temperaturabhängig gemessen und verschiedene Methoden der Datenanalyse vorgestellt und diskutiert. Im letzten und gleichzeitig komplexesten Problem dieser Arbeit geht es um die Kombination der verschiedenen Methoden zur simultanen Bestimmung von dynamischen und strukturellen Eigenschaften eines kompletten Katalysezyklus. Die Untersuchung erfolgte mittels zeitaufgelöster Resonanz-Raman-Spektroskopie, deren Spektren nicht nur einen Überblick über den zeitlichen Verlauf des Prozesses liefern, sondern die Identifizierung intermediär gebildeter Zustände erlauben. Für die Analyse wurde zusätzlich ein mathematisches Verfahren der simultanen Bestimmung von Komponenten und ihrer Kinetik angewandt und die Ergebnisse denen der vorangegangen Methoden gegenübergestellt.
The aim of this thesis is the use of chemometric methods for the analysis of complex data sets that are generated by Raman spectroscopy. On the basis of selected examples, suitable strategies for deciphering the chemical and physical information of a biological or chemical system from experimental spectra are presented and evaluated. Common objective of the methods is the determination of the \textit{Eigen}-spectra of the species of the respective system (component analysis). The component spectra obtained in this way constitute the basis for analyzing the distribution of the species involved as well as their structural, thermodynamic, or kinetic relationships. The present work documents the manifold of applications of the quantitative spectra analysis and its computer-assisted automation. Using the example of quality control of coffee beans, it is shown that for qualitatively and quantitatively distinguishing between two species, a single spectral marker may be sufficient, even if its nature is unknown. Through the use of algorithmic methods the control of statistically meaningful number of samples could be accelerated. However, Raman spectroscopy allows not only the differentiation of species, but also their structural characterization. The example of the biological photoreceptor rhodopsin demonstrates the possibilities for structure analysis in structural variable systems. The determination of the component spectra of the species involved in an equilibrium then allows for evaluating thermodynamic parameters. This is demonstrated on the basis of Raman spectra of a prototypical phytochrome, measured as a function of the temperature. Various methods of data analysis are presented to determine thermodynamic variables of the system. The last and most complex problem of this work refers to the simultaneous determination of the dynamic and structural properties of a complete catalytic cycle. The investigation was carried out by means of time-resolved resonance Raman spectroscopy, the spectra of which not only provide an overview of the temporal evolution of the process but also allow for identifying intermediately formed states. For the analysis, a mathematical method for the simultaneous determination of components and their kinetics was applied and the results compared with the previous methods.
