dc.description.abstractIm ersten Teil dieser Arbeit stand die enantioselektive Darstellung von Isoxazolinen im Vordergrund. Der synthetische Ansatz beinhaltete die asymmetrisch katalysierte 1,3-dipolare Cycloaddition von Nitronen an Alkine. Hierfür wurden sowohl elektronenarme mono- und bifunktionelle als auch elektronenreiche Dreifachbindungssysteme, in Kombination mit elektronenreichen und elektronenarmen Nitronen herangezogen. Als asymmetrische Induktoren standen sowohl klassische Lewissäure/Ligand-Kombinationen als auch Organokatalysatoren im Fokus. Mittels Organokatalyse konnte erstmals eine Enantioselektivität erzielt werden, die zugleich mit 67% ee auch die erste nennenswerte stereoselektive 1,3-dipolare Cycloaddition eines Nitrons mit einem Alkin darstellt. Der zweite Teil dieser Arbeit befaßt sich vornehmlich mit Folgeprodukten, die sich aus einer thermischen, reduktiven und oxidativen N-O-Bindungsspaltung sowie einer palladiumkatalysierten Umlagerung von Isoxazolinen ergäben. So lag im Zusammenhang mit der thermischen N-O-Bindungsspaltung von Isoxazolinen das Hauptaugenmerk vor allem in einem einfachen und effizienten Zugang zu Lamellarinderivaten. Die reduktive N-O-Bindungsspaltung zur Darstellung von beta-Aminocarbonylverbindungen wurde zunächst durch Umsetzung der Isoxazoline mit SmI2 in Methanol und folgend erstmals erfolgreich mittels Zn in Anwesenheit von HCl durchgeführt. Desweitern wurde im Rahmen dieser Arbeit entdeckt, daß C3-H-substituierte Isoxazoline in Anwesenheit von Pd(PPh3)4 und HSiR3 zu den entsprechenden Enaminonen umlagern. Optimierungen hinsichtlich der eingesetzten Silane und Reaktionsbedingungen konnten erreicht und ein möglicher Katalysecyclus aufgestellt werden. Durch Versetzen von bicyclischen Isoxazolinen mit mCPBA wurde eine neuartige Darstellung von oxidativen N-O-Bindungsspaltungsprodukten von Isoxazolinen entwickelt. Je nach Substitutionsmuster erfolgt diese unter einfacher oder auch zweifacher Ringspaltung. Dadurch werden Nitrone der 2. Generation bzw. primäre Nitrosoverbindungen in sehr guten Ausbeuten zugänglich, welche sich in Form der dimeren Diazodioxidverbindung stabilisieren. Strukturelle, stereochemische und quantenmechanische Betrachtungen der Reaktion haben einen plausiblen Reaktionsmechanismus ergeben. Die Darstellung der stark aktivierten alpha,beta-ungesättigten-omega-Nitroso- und Nitrotricarbonylverbindungen eröffneten einen neuen und eleganten Weg zur Synthese von Pyridazin-3-onen und Acylhydrazinderivaten, der im Rahmen dieser Arbeit erfolgreich aufgezeigt wurde. Ein möglicher Reaktionsmechanismus und Ansätze zur Folgechemie ausgehend von Acylhydrazinen werden vorgestellt.de
dc.description.abstractThe first part of the presented work deals with the enantioselective synthesis of isoxazolines. The synthetic approach involves the asymmetrically catalysed 1,3-dipolar cycloaddition of nitrones to acetylenes. Therefore electrondeficient mono- and bifunctionalysed as well as electronrich triplebond systems, in combination with electronrich and electronpoor nitrones were taken into account. As asymmetric inductors classic lewisacid/ligand combinations as well as organocatalysts were used. An enantioselectivity of 67% ee representing the first noteworthy stereoselective 1,3-dipolar cycloaddition of a nitron with an acetylene, was achieved by means of organocatalysis. Furthermore isoxazolines were used as synthons in order to achieve a widespread chemistry leading to a extended structure variety of compounds with different overall oxidation states. The thermal and substituent depended stability of several synthesised isoxazolines has been tested in order to give an access to interesting heterocycles. Especially the transformation into tetrahydroisochinoline fused pyrrole derivatives was used as synthetic approach to Lamellarine derivatives. The synthesis of beta-amino-carbonyl compounds starting from isoxazolines through a reductive procedure has been tested at first with SmI2/methanol and finally was achieved by means of Zn in presence of an acid. A novel rearrangement of C3-H substituted isoxazolines to enaminones was observed by use of Pd(PPH3)4/HSiR3. Reaction optimisations regarding the used silanes and reaction conditions could be achieved and a mechanism for the observed transformation is proposed. By treatment of bicyclic isoxazolines with mcpba an entry to novel oxidation products was accomplished. Depending on the substitution pattern nitrones of the 2nd generation or alpha,beta-unsaturated-omega-nitroso compounds are furnished. The structural requirements, stereochemical outcome and quantumchemical calculations have revealed a probable mechanism. The strongly activated doublebond of the synthesised alpha,beta-unsaturated-tricarbonyl compounds gives an efficient access to pyridazin-3-ones and acylhydrazines which has been opened within this work. A probable mechanism and preliminary studies on the synthetic potential of the synthesised acylhydrazines are presented.en
