Bestimmung von Tocopheroloxidationsprodukten im Humanserum

dc.description.abstractDie vorliegende Arbeit beschäftigt sich mit den Tocopherolen (Vitamin E) und ihren Oxidationsprodukten, wobei der Schwerpunkt bei der Entwicklung einer Methode zur Bestimmung von Tocopheroloxidationsprodukten in Humanserumproben lag. Die Methodenentwicklung konzentrierte sich auf die Bestimmung des alpha-Tocopherolchinons (alpha-TQ) als bedeutendes Oxidationsprodukt des wirksamsten und im Serum dominierenden Vertreters der Vitamin-E-Gruppe, des alpha-Tocopherols. Weiterhin wurde die Möglichkeit der Bestimmung der beta-, gamma- und delta-Tocopherolchinone (beta-, gamma-, delta-TQ) sowie der entsprechenden Tocopherolhydrochinone (alpha-, beta-, gamma-, delta-THQ) getestet. Hierzu erfolgte zunächst eine Synthese der Referenzsubstanzen mittels gängiger Syntheseverfahren und anschließend eine Charakterisierung der Syntheseprodukte mittels verschiedener analytischer Verfahren. Dabei wurden Daten zu deren chromatographischen Eigenschaften in der lipophilen Gelchromatographie, der Gaschromatographie (GC) und der Hochleistungs-Flüssigchromatographie (HPLC) sowie zu den Massenspektren der Substanzen erarbeitet, die zumindest für beta-, gamma- und delta-TQ und THQ in der Literatur bisher nur sehr eingeschränkt bzw. gar nicht verfügbar sind. Die Untersuchungen zeigten, dass eine direkte Bestimmung der Tocopherolhydrochinone im Serum aufgrund deren äußerst hoher Oxidationsempfindlichkeit nur in frischen Serumproben sinnvoll sowie nur mit sehr hohem Aufwand bei der Probenvorbereitung möglich ist. Für beta-, gamma- und delta-TQ wurden bei der Extraktion aus dem Serum unbefriedigende Wiederfindungsraten erzielt, deren Ursache vermutlich in einer Bildung von Addukten mit Substanzen wie Glutathion oder Proteinen liegt, wodurch diese Tocopherolchinone zumindest partiell der Bestimmung entzogen werden. Dagegen ist eine Bildung solcher Produkte beim vollständig methylierten alpha-TQ nicht möglich, so dass für diese Substanz hervorragende Wiederfindungsraten erzielt wurden. Die entwickelte Bestimmungsmethode ermöglicht eine zuverlässige Analytik von alpha-TQ in Serumproben, wobei eventuell ebenfalls in den Proben enthaltenes alpha-THQ als alpha-TQ miterfasst wird. Die zweistufige Probenvorbereitung kombiniert eine Lipidextraktion mit einem säulenchromatographischen Reinigungsschritt an Kieselgel. Zur Messung wurde eine neuartige RP-HPLC-Methode erarbeitet, bei der durch eine Nachsäulenderivatisierung mit Hilfe eines Photoreaktors eine selektive und sensitive Bestimmung der Tocopherolchinone mittels Fluoreszenzdetektion ermöglicht wird. Die Nachweisgrenze der Messmethode für alpha-TQ liegt bei ca. 320 pg. Weitere Bestandteile der Arbeit waren die Anwendung der entwickelten Methode zur Bestimmung der alpha-Tocopherolchinon-Gehalte für eine Auswahl von Serumproben des Bundes-Gesundheitssurveys 1998 sowie eine Auswertung und Interpretation der erzielten Ergebnisse. Ebenso erfolgte eine Auswertung der in einer größeren Stichprobe von Seren des Surveys bestimmten Gehalte an alpha-, beta-, gamma- und delta-Tocopherol. Dabei wurden besonders die Unterschiede zwischen den Anwendern von Vitamin-E-Präparaten und einer Kontrollgruppe von Personen ohne Vitamin-E-Einnahme untersucht.de
dc.description.abstractThe central topic of this thesis are the tocopherols (vitamin E) and their oxidation products, with special emphasis on the development of a method for determination of tocopherol oxidation products in human serum samples. Method development focused on determination of alpha-tocopherolquinone (alpha-TQ), an important oxidation product of alpha-tocopherol, which is the vitamin E compound with the highest biological activity and also the most abundant vitamin E compound in serum. Additionally, the possibilities for determination of beta-, gamma- and delta-tocopherolquinones (beta-, gamma-, delta-TQ) and the respective tocopherolhydroquinones (alpha-, beta-, gamma-, delta-THQ) were evaluated. TQ and THQ reference substances were synthesized by standard synthesis procedures and analytical data for the chromatographic properties of these compounds in lipophilic gel chromatography, gas chromatography (GC) and high performance liquid chromatography (HPLC) as well as their mass spectra were acquired, that were mostly not documented in literature so far. The further research revealed, that because of their high susceptibility to oxidation, a direct determination of THQ´s would only be useful in fresh serum samples and even then would require extensive efforts to prevent oxidation during sample workup. Also, recoveries for beta;-, gamma;- and delta-TQ from serum were unsatisfactory, probably due to formation of nucleophilic adducts with compounds like glutathione or proteins, which prevent complete isolation of these TQ´s from the serum matrix. On the other hand, excellent recoveries were obtained for the fully methylated alpha-TQ, that is not able to form such adducts. The finally developed method allows a reliable determination of alpha-TQ in human serum samples, where possibly further contained alpha-THQ is also measured as alpha-TQ. The two-step sample preparation procedure combines a lipid extraction step with a clean up step on a little silica gel column. For measurement, a novel RP-HPLC method has been developed, that allows selective and sensitive fluorescence detection of tocopherolquinones after post-column derivatization by a photoreactor. The limit of detection for alpha-TQ is about 320 pg. Further parts of this work were the application of the developed method to the determination of alpha-TQ in a subsample of serum samples from the German National Health Survey 1998 and an evaluation and discussion of the results obtained. Additionally, data for alpha-, beta-, gamma- and delta-tocopherol concentrations in serum, measured by a standard laboratory procedure in a larger subsample of the German National Health Survey 1998, are discussed with special emphasis on differences between users of tocopherol supplements and a control group of no-drug users.en
