Makrocyclische C2-symmetrische Kohlenhydrate und Synthese von (+)-7a-epi-7-Deoxycasuarin durch Olefinmetathese

dc.description.abstractDie Kreuzmetathese (CM) konnte sich mit der Entwicklung neuer Katalysatoren als intermolekulare Standard-C-C-Verknüpfungsmethode etablieren. Die hohe Effizienz der CM von akzeptorsubstituierten Olefinen mit elektronisch neutralen Alkenen bei hoher E/Z-Selektivität erlaubt die Verlängerung der Kohlenstoffkette um beliebig viele C-Atome. Im ersten Teil der vorliegenden Arbeit wurde die Cyclodimerisierung von geeignet substituierten Kohlenhydraten über eine Tandemreaktion aus Kreuzmetathese und Ringschlussmetathese beschrieben. Die 1,6-substituierten Zucker ergaben bei den Cyclodimerisierungen in guten Ausbeuten von 41-65% die makrocyclischen C2-symmetrischen Cyclodimere in einer Kopf-Schwanz-selektiven Reaktion als reine E-Isomere. Die Ausbeute an Cyclodimer nahm dabei mit zunehmender Ringgröße ab. Es ließen sich sowohl Benzyl- und Acetyl-geschützte C-Mannoside bzw. C-Glucoside, als auch Benzyl-geschützte beta- und alpha-Anomere von O-Glucosiden umsetzen. In wenigen Fällen konnten kleine Mengen des Cyclotrimers isoliert werden. Bei Zuckerderivaten mit O-allylischer Doppelbindung wurde Isomerisierung zum Enolether und keine Cyclodimerbildung beobachtet. 1,2-, 1,3- und 1,4-substituierte C-Glucoside wurden ebenfalls in einer Metathese zur Cyclodimerisierung umgesetzt. Hierbei wurden acyclische Oligomere als Produkte erhalten, und es konnte nur in einem Fall eine kleine Menge Cyclodimer (11%) isoliert werden. Mit einem modifizierten Konzept wurden symmetrische Platzhalter für Peptide, sogenannte Peptidmimetika, und Zucker miteinander in einer Metathese cyclisch verknüpft. Die cyclischen Glycopeptidmimetika konnten in recht guten Ausbeuten von 32-44% erhalten werden. Im zweiten Teil der vorliegenden Arbeit wurden Kreuzmetathesen von verschiedenen Vinylpyrrolidinen mit akzeptorsubstituierten Olefinen und mit einem Allylalkohol vorgestellt, die in guten Ausbeuten stereoselektiv die E-konfigurierten CM-Produkte liefern. Nachfolgend wurde eine Reaktionssequenz aus CM von Vinylpyrrolidin (oder –derivat), asymmetrischer Dihydroxylierung und cyclisierender, reduktiver Aminierung zur Darstellung von polyhydroxylierten 3-Hydroxymethylpyrrolizidin-Alkaloiden entwickelt. Mit dieser Syntheseroute wurde (+)-7a-epi-7-Deoxycasuarin (122) ausgehend von kommerziell erhältlichem L-4-Hydroxyprolin in neun Stufen und einer Gesamtausbeute von 15% synthetisiert. Analog wurde das polyhydroxylierte Pyrrolizidin 144 in neun Stufen und einer Gesamtausbeute von 11% erhalten.de
dc.description.abstractWith the development of new catalysts cross metathesis (CM) was established as a intermolecular standard C-C-coupling method. The high efficiency of cross metathesis of acceptor-substituted olefins with electronic neutral alkenes by high E/Z-selectivity allows elongation of carbon chains by arbitrary C-atoms. In the first part of this thesis cyclodimerisation of appropriate substituted carbohydrates by a tandem reaction of cross metathesis and ring closing metathesis was described. In these cyclodimerisations 1,6-substituted carbohydrates gave good yields of 41-65% macrocyclic C2-symmetric cyclodimers as pure E-isomers in a head-tail-selective reaction. Thereby the yield decreases with increasing ring size. It was possible to convert benzyl- and acetyl-protected C-mannosides and C-glucosides respectively as well as benzyl-protected beta- and alpha-anomers of O-glucosides. In few cases small amounts of cyclotrimer could be isolated. On sugar derivatives with O-allylic doublebond isomerisation to the enolether and no formation of cylodimers was observed. 1,2-, 1,3- and 1,4-substituted C-glucosides were also converted in an olefin metathesis for cyclodimerisation. Thereby acyclic oligomers as products were obtained and only in one case a small amount of cyclodimer (11%) could be isolated. With a modified concept symmetric placeholder for peptides, so-called peptidomimetics, and sugars were connected with each other to a cycle by olefin metathesis. The cyclic glycopeptidomimetics could be obtained in quite good yields of 32-44%. In the second part of this thesis cross metathesis reactions of different vinylpyrrolidines with acceptor-substituted olefins and an allylic alcohol were presented, they afford cross metathesis products in good yields and complete E-stereoselectivity. Subsequent to this a reaction sequence of CM of vinylpyrrolidine (or –derivative), asymmetric dihydroxylation and cyclic reductive amination was developed for the construction of polyhydroxylated 3-hydroxymethylpyrrolizidine-alkaloids. Using this concept (+)-7a-epi-7-deoxycasuarine (122) was synthesised in nine steps with an overall yield of 15% starting from commercially available L-4-Hydroxyprolin. Analog the polyhydroxylated pyrrolizidine 144 was obtained in nine steps with an overall yield of 11%.en
