dc.description.abstractIn der vorliegenden Arbeit wurden die antioxidative Aktivität sowie die immunmodulierende Kapazität von Zellwänden und Zellwandbestandteilen der Bierhefe Saccharomyces cerevisiae untersucht. Die Zellwände von Saccharomyces-Hefen bestehen aus (1→3),(1→6)-β-D-Glucan, Mannanen, Proteinen und Lipiden. Als potentieller Immunmodulator ist vor allem das (1→3),(1→6)-β-D-Glucan von Interesse. Vor kurzem konnte für dieses Glucan zusätzlich eine antioxidative Wirkung nachgewiesen werden [KOGAN et al., 2005]. Obwohl das Hauptinteresse der Untersuchungen dem β-Glucan galt, sollte der denkbare Beitrag anderer Zellwandkomponenten zur bioaktiven Wirkung von Hefezellwänden berücksichtigt werden. Da die Zellwandzusammensetzung und die damit verbundene Bioaktivität von den Wachstumsbedingungen der Hefe abhängen, wurde Hefe auf verschiedenen Medien mit unterschiedlichen Kohlenstoffquellen (Bierwürze, Hefe-Pepton-Glucose- und Hefe-Pepton-Galactose-Medium) herangezogen. Anschließend wurden mit einem speziellen Verfahren die Komponenten selektiv aus den Hefezellwänden entfernt, so dass verschiedene Fraktionen mit unterschiedlichen Konzentrationen an Zellwandbestandteilen gewonnen wurden. Nach der Aufarbeitung wurde der Gehalt der einzelnen Fraktionen an β-Glucan, Mannan und Proteinen bestimmt. Die immunmodulierende Wirkung wurde aus der Cytokinsekretion mononuklearer peripherer Blutzellen (PBMC) mittels ELISA gemessen. Die antioxidative Aktivität wurde mittels Elektronenspinresonanzspektroskopie (ESR), der „Trolox Equivalent Antioxidant Capacity“-Methode (TEAC) und einem „amperometrischen Biosensor“ bestimmt. Die Ergebnisse zeigen, dass die Zusammensetzung der Hefezellwände in hohem Maße vom Wachstumsmedium und insbesondere von der Kohlenstoffquelle abhängt. Auch auf die antioxidative Aktivität haben Wachstumsmedium und Kohlenstoffquelle einen großen Einfluss. Die in der Literatur beschriebene antioxidative Wirkung von hochangereichertem β-Glucan aus S. cerevisiae konnte durch die Messungen nicht bestätigt werden. Dagegen scheinen vor allem die in der Hefezellwand enthaltenen Proteine eine starke antioxidative Wirkung besitzen. Als antioxidativ wirkende Gruppen kämen dabei aromatische Seitenketten und freie Thiole aus denaturierten Proteinen in Frage, wobei die Intensität der antioxidativen Aktivität wahrscheinlich von der Zugänglichkeit zu diesen Gruppen abhängt. Die Untersuchung der immunmodulierenden Wirkung ergab eine deutliche Stimulation der PBMC durch die gewonnenen Hefezellwandfraktionen. Dabei gab es erhebliche Unterschiede zwischen den Fraktionen, was beweist, dass verschiedene Zellwandkomponenten zur immunstimulierenden Wirkung beitragen und nicht alleine das (1→3),(1→6)-β-D-Glucan dafür verantwortlich ist.de
dc.description.abstractIn the present work the antioxidative activity and the immunomodulating capacity of Saccharomyces cerevisiae cell walls and cell wall components were examined. The cell walls of this yeast consist of (1→3),(1→6)-β-D-glucan, mannans, proteins and lipids. As potential immunomodulator (1→3), (1→6)-β-D-glucan is of great interest. Recently, these glucans were found to show a certain antioxidative activity [KOGAN et al., 2005]. Although the main interest of the investigations was focussed on β-glucan, the possible contribution of the other cell wall components to the cell walls’ bioactivity should be considered, too. Since cell wall composition and bioactivity strongly depend on growth conditions of the yeast, different growth media with different carbon sources (wort, yeast-peptone-glucose and yeast-peptone-galactose-medium) were applied. Subsequently, the cell wall components were removed selectively from the yeast cell walls with a special extraction procedure, thus, different call wall fractions with different concentrations of cell wall components were gained. After fractionation the content of β-glucans, mannans and proteins in the different cell wall fractions was determined. The immunomodulating effect was measured from cytokine secretion of peripheral mononuclear blood cells (PBMC) by ELISA. The antioxidative activity was analysed by electron spin resonance spectroscopy (ESR), by the “Trolox Equivalent Antioxidant Capacity“ (TEAC) assay and by an “amperometric biosensor“. The results show that the composition of the yeast cell walls depends considerably on growth medium and carbon source. Growth medium and carbon source also influence antioxidative activity, greatly. The antioxidative effect of highly purified β-glucan from Saccharomyces cerevisiae, as described in literature, could not be confirmed in the present study. However, there is evidence that the cell wall proteins have a strong antioxidative effect. More precisely, aromatic side chains and maybe thiol groups of the proteins seem to act as antioxidants whereas the intensity of the antioxidative activity apparently depends on the accessibility of these groups. The investigations on the immunomodulating effect show a clear stimulation of the PBMC by the yeast cell wall fractions. There were significant differences between the fractions, which proves that different cell wall components contribute to the immunostimulating effect. Accordingly, (1→3), (1→6)-β-D-glucan is not responsible alone for the observed immunomodulating capability of yeast cell walls.en
