Aufnahme und Toxizität von Silbernanopartikeln im Intestinalmodell Caco-2

dc.description.abstractDie Nanotechnologie gilt vor allem in der Industrie als eine wichtige Zukunftstechnologie. Zu den Nanoobjekten zählen auch Nanopartikel, die in allen drei Dimensionen eine Ausdehnung von 1 bis 100 nm haben. Ausgangspunkt dieser Arbeit war eine Stellungnahme der Europäischen Lebensmittelsicherheitsbehörde (EFSA), die sich aufgrund fehlender Daten nicht in der Lage sah, die Bewertung von kolloidalem Silber als Nahrungsergänzungsmittel vorzunehmen. Zur genaueren Charakterisierung der Auswirkungen von Tensid-beschichteten Silbernanopartikeln auf die gastrointestinale Barriere wurde in dieser Arbeit die Aufnahme über das humane Dünndarmepithel mittels Caco-2-Zellmodell in der Transwell®-Technik sowie elektronenmikroskopischer Bildgebung untersucht. Es zeigte sich, dass die Silbernanopartikel in nicht zytotoxischen Konzentrationen das Epithel nicht in größerer Menge passieren oder von Enterozyten aufgenommen werden. Die beobachtete Zytotoxizität in höheren Nanopartikelkonzentrationen kann von mehreren Faktoren beeinflusst werden. Einen deutlichen Einfluss auf die Zytotoxizität zeigten die Silberionenfreisetzung und die Größe der Partikel. Die Zusammensetzung des Mediums zeigte nur einen geringen Einfluss auf die Zytotoxizität. Insgesamt deutete sich ein sehr komplexes Zusammenspiel der verschiedenen Nanopartikel-, Assay- und Zelleigenschaften an. Die Versuche zur Trennung der Einflüsse von partikulärer Form und freigesetzten Ionen mittels Ultrazentrifugation und Dialyse zeigten, dass die Silberionen eine entscheidende Rolle für die Wirkung von Silbernanopartikeln spielen. Jedoch konnte nicht endgültig zwischen Partikeln und Ionen unterschieden werden, da es sich um einen kontinuierlichen Prozess und ein kompliziertes Wechselspiel beider Partner handelt. Zur Untersuchung des zellulären Wirkungsmechanismus der Tensid-beschichteten Silbernanopartikel wurden weiterhin Assays zur Messung des oxidativen Stresses, Glutathiongehaltes, Calciumeinstroms, der mitochondrialen Depolarisation und Impedanz sowie Micro-Array und PCR-Analysen verwendet. Diese Analysen zeigten, dass die Wirkung von Silbernanopartikeln und Ionen auf Intestinalzellen vermutlich über die Wechselwirkung mit membranständigen Proteinen, über das Eindringen von Ionen in die Zellen und über oxidativen Stress vermittelt wurde. Diese nahmen Einfluss auf die Calciumhomöostase und verschiedene Stoffwechselwege, die unter anderem über die Fokalen Adhäsionen (FAK), Integrine (ILK) und Zell-Zell-Verbindungen vermittelt werden sowie Kinase-vermittelte Signalwege (ERK / MAPK). Sie induzierten verschiedene Transkriptionsfaktoren und lösten somit eine zelluläre Reaktion auf reaktive Sauerstoffspezies und den Calciumeinstrom aus. Zudem waren Auswirkungen auf das Zytoskelett sowie die Zell-Zell- und Zell-Matrix-Kontakte der Zellen zu beobachten. In Caco 2-Zellen kam es für höhere Konzentrationen schließlich zur Nekrose. Für die Risikobewertung wird zum einen die tatsächliche Exposition des Menschen mit Silbernanopartikeln und zum anderen die Betrachtung der Eigenschaften und Auswirkungen dieser benötigt. Umfassende Daten zur Exposition liegen noch nicht vor, jedoch ist aufgrund der steigenden Verwendung von Silbernanopartikeln in Zukunft mit einem vermehrten Vorkommen zu rechnen. Die Betrachtung der Auswirkungen von Tensid-beschichteten Silbernanopartikeln auf Intestinalzellen in dieser Arbeit zeigt, dass diese nicht aktiv aufgenommen werden. Trotzdem sind eine konzentrationsabhängige zytotoxische Wirkung sowie deutliche Auswirkungen auf den zellulären Stoffwechsel der Intestinalzellen zu erkennen. Die Ursachen der von einer Aufnahme unabhängigen molekularen Effekte auf das Dünndarmepithel bedürfen weiterer Untersuchungen.de
dc.description.abstractIn industry, nanotechnology is regarded as the technology of the future. Nanoobjects also include nanoparticles, which range in size from 1 to 100 nm in all three dimensions. The starting point for this work was a statement of the European Food and Safety Authority (EFSA), which pointed out that they are incapable of assessing the risk of colloidal silver supplements, due to a lack of data. The uptake and toxicity of nanomaterials is important as a basis for risk assessment. After oral administration the intestine is the first barrier for the resorption. For more precise characterization of the effects of silver nanoparticles on the gastrointestinal barrier, the Caco 2 cell line was used. For the characterization of the uptake of surfactant coated silver nanoparticles the Transwell® technique and electron microscopy were applyed. It has been shown that the surfactant coated silver nanoparticles used do not pass the gastrointestinal epithelium in great quantities, when they are used in non-cytotoxic concentrations. The observable cytotoxicity for higher concentrations of nanoparticles can be influenced by several factors. A clear influence on the cytotoxicity was detectable for the silver ion release rate and the particle size. The composition of the cell culture medium was minor for the surfactant coated silver nanoparticles. The influence of the different nanoparticle coatings, surfactant, tannic acid, PVP and peptides, was beyond recognition. This suggests complex interactions of the different properties of nanoparticles, assays and cells. The attempts to separate the influence of the silver particles from the released silver ions, by ultracentrifugation and dialysis, showed that the ions play an important role for the effect of the silver nanoparticles on cells. However it was not possible to definitely separate the effects of particles and ions, because of the extensive interplay between the particles and ions, especially in complex solutions like cell culture media. To investigate the molecular mechanisms of the surfactant coated silver nanoparticles, different assays were used to measure the oxidative stress, glutathion content, calcium flux, mitochondrial depolarization, impedance, micro array and PCR analysis. These examinations showed an effect of the silver nanoparticles and silver ions on the intestinal cells, which was probably mediated by proteins located in the cell membrane, the invasion on ions, or by oxidative stress. This exerted influence on the calcium homeostasis and different metabolic pathways like Focal Adhesion Kinase (FAK), Integrin Linked Kinase (ILK), Extracellular Signal Regulated Kinases / Mitogen-Activated Protein Kinases (ERK / MAPK), Integrin, Epithel Adherens Junction und Tight Junction Signaling. Furthermore several transcription factors were induced. There were also consequences for the cellular cytoskeleton as well as for the cell-cell contacts and cell-matrix contacts of the cells. High concentrations caused necrosis in Caco-2 cells. For risk assessment is on the one hand the oral exposure of silver nanoparticles and on the other hand the consideration of toxicity nessesary. Complete exposition data are not available, but the applications of silver nanoparticles are rising and so will the exposition. The data of this PhD thesis showed that surfactant coated silver nanoparticles are not actively taken up by intestinal cells. Although a clear influence on the metabolic activity and pathways were detectable. Nevertheless, the effect of silver nanoparticles on intestinal cells requires further study.en
