dc.description.abstractSynthese, Charakterisierung und Eigenschaften von Oktaaza- und Tetraazamakrozyklen In dieser Arbeit wurden Wege für die Synthese von neuen Azakronenethern aufgezeigt, die als Liganden für die Komplexierung von z.B. Gadolinium dienen können, das in der Magnet-Resonanz-Tomographie vielfach Anwendung findet. Eine oktaazamakrozyklische Ligandenvorstufe, die an den Stickstoffatomen durch 4-Nitrophenylsulfonyl geschützt vorliegt konnten mit Templatunterstützung über eine [1+1]-Zyklisierung zweier ungleich großer Fragmente durch Cäsiumcarbonat über eine 12-stufige Synthese dargestellt und charakterisiert werden. Dabei wurde die Verbindung 1,13-Dibenzyl-4,7,10,16,19,22-hexakis(4-nosyl)-1,4,7,10,13,16,19,22-oktaazazyklotetrakosan (20) ausgehend von Diethylentriamin und Aminoethanol gewonnen. Die Darstellung der Zwischenstufen N,N‘-(4-Nosyl)-3-benzyl-3-aza-1,5-pentandiamin (4), 1-(Methansulfonyl)-3-(4-Nosyl)-5-[2(tetrahydropyran-2-yloxy)]-3-aza-pentan (14), 1,4,10,13-Tetrakis(4-nosyl)-7-benzyl-1,4,7,10,13-pentaaza-1,13-bis[2-(tetrahydro-pyran-2-yloxy)-ethyl]-tridekan (17), 1,4,10,13-Tetrakis(4-nosyl)-7-benzyl-1,4,7,10,13-pentaaza-1,13-bis[2-(hydroxy)-ethyl]-tridekan (18) wie auch 9-Benzyl-3,6,12,15-tetrakis(4-nosyl)-3,6,9,12,15-pentaazaheptadekan-1,17-diyldimethansulfonsäureester (19) wurden ebenfalls beschrieben. Die Molekülstrukturen der Verbindungen 4-Benzyldiethylentriamin-Dihydrochlorid (3∙2HCl), 4-Methyl-N,N-bis(2-(4-methylbenzensulfonamido)ethyl)benzensulfonamid (26) und 4-Benzyl-1,7-bis-(4-tosyl)-diethylentriamin (27) konnten über eine Röntgenstrukturanalyse aufgeklärt werden. Über eine [2+2]-Zyklisierung zweier gleichgroßer Fragmente (4 und 2,2'-(4-Nitrobenzenensulfonamido)bis(ethan-2,1-diyl)dimethansulfonsäureester (16)) konnte Verbindung 20 ebenfalls, wenn auch mit geringerer Ausbeute, gewonnen werden. Auf diesem Wege ließen sich auch die Bn/Ns/Ts- gemischt geschützte Verbindung 1,13-Dibenzyl-4,10,16,22-tetrakis(4-nosyl)-7,19-ditosyl-1,4,7,10,13,16,19,22-oktaazazyklotetrakosan (41) und die Bn/Ts-Verbindung 1,13-Dibenzyl-4,7,10,16,19,22-hexatosyl-1,4,7,10,13,16,19,22-oktaazazyklotetrakosan (38) darstellen. Diese Reaktion lieferte als Produkt gleichzeitig die aus einer [1+1] hervorgegangenen Zyklisierung entstandenen Verbindungen 1-Benzyl-4,7,10-tritosyl-1,4,7,10-tetraazazyklododekan (35), 1-Benzyl-4,7,10-tris(4-nosyl)-1,4,7,10-tetraazazyklododekan (39) und 1-Benzyl-4,10-bis(4-nosyl)-7-tosyl-1,4,7,10-tetraazazyklododekan (40). Von Verbindung 35 konnte die Molekülstruktur gewonnen werden. Die analog zu 40 aufgebaute, an N10 Heptyl substituierte, Verbindung konnte ebenfalls mit ihrer Molekülstruktur gewonnen werden. Die Verbindung 27 reagierte mit Uranylnitrat als potenziellem Templat nicht über eine Zyklisierung, sondern redoxchemisch unter Bildung von Uranoxid zu N,N-bis(2-(4-methylbenzensulfonamido)ethyl)formamid (43). Die Massenspektroskopie gibt Hinweise auf Mesomere und Tautomere. Zwei Mono-N-funktionalisierte, dreifach mit Essigsäure alkylierte DO3A-Derivate 10-(2-Propenyl)-1,4,7,10-tetraazazyklododekan-1,4,7-triessigsäure (48) und 10-(3-Butenyl)-1,4,7,10-tetraazazyklododekan-1,4,7-triessigsäure (55) wurden dargestellt und mit den Metallionen Samarium, Lutetium und Thulium zu den entsprechenden Komplexen [10-(2-Propenyl)-1,4,7,10-tetraazazyklododekan-1,4,7-triacetato]samarium (49), [10-(2-Propenyl)-1,4,7,10-tetraazazyklododekan-1,4,7-triacetato]lutetium (50), [10-(2-Propenyl)-1,4,7,10-tetraazazyklododekan-1,4,7-triacetato]thulium (51) und [10-(3-Butenyl)-1,4,7,10-tetraazazyklododekan-1,4,7-triacetato]thulium (56) umgesetzt. Zwei Tetra-N-funktionalisierte Cyclen-Derivate wurden dargestellt. Das 1,4,7,10-Tetra-2-propenyl-1,4,7,10-tetraazazyklododekan (57) wurde mit den Metallen Nickel (58), Kupfer (61), Thulium (62), Eisen (63) Kobalt (64), Lithium (60) und Rhodium (65) unter Komplexbildung umgesetzt. Der Komplex 58 lieferte nach Umsetzung mit Kupferchlorid einen Olefinkomplex (59), dessen Struktur aufgeklärt werden konnte, wie auch die Strukturen von 58 und 61. Der Ligand 1,4,7,10-Tetra(3-butenyl)-1,4,7,10-tetraazazyklododekan (66) lieferte nach Umsetzung mit Nickel, Thulium und Eisen die Verbindungen 67, 68 und 69. Die Struktur der Verbindung 67 konnte ebenfalls aufgeklärt werden. In den jeweiligen Strukturen befinden sich die Metallatome durchwegs pyramidal umgeben im Abstand von 0.5 und 0.8 Å der von den 4N-Atomen aufgespannten Ebene. Ein Tri-N-funktionalisierter Ligand, 1,4,7-Tri-(2-pyridyl)-1,4,7,10-tetraazazyklododekan (70) wurde gewonnen und mit Thuliumchlorid zur Verbindung 71 umgesetzt. Die dargestellten Thulium-Komplexverbindungen zeigen fluoreszierende Eigenschaften, die durch Aufnahme von Fluoreszenzsspektren untersucht und miteinander verglichen wurden. Von den Eisen-, Kobalt- und Rhodium-Verbindungen wurden Zyklovoltammogramme in Acetonitril zur Analyse der redoxchemischen Eigenschaften aufgenommen.de
dc.description.abstractSynthesis, characterization and properties of octaaza- and tetraazamacrocycles In this thesis different routes for the synthesis of new aza crown ethers were proposed. These could act as ligands for the complexation of gadolinium that is used frequently in magnetic resonance imaging techniques. A six-fold 4-nitrobenzenesulfonyl protected octaazamacrocyclic precursor of a ligand was synthesized over a [1+1]-cyclization from two unequal fragments by assistance of cesiumcarbonate as a template via 12 steps and subse-quently characterized. Starting with diethylenetriamine and ethanolamine the compound 1,13-dibenzyl-4,7,10,16,19,22-hexakis(4-nitrobenzenesulfonyl)-1,4,7,10,13,16,19,22-octaazacyclotetracosane (20) was isolated. The preparation of the intermediates N,N‘-(4-nitrobenzenesulfonyl)-3-benzyl-3-aza-1,5-pentandiamine (4), 1-(methanesulfonyl)-3-(4-nitrobenzenesulfonyl)-5-[2(tetrahydropyran-2-yloxy)]-3-aza-pentane (14), 1,4,10,13-tetrakis(4-nitrobenzenesulfonyl)-7-benzyl-1,4,7,10,13-pentaaza-1,13-bis[2-(tetrahydro-pyrane-2-yloxy)-ethyl]-tridecane (17), 1,4,10,13-tetrakis(4-nitrobenzenesulfonyl)-7-benzyl-1,4,7,10,13-pentaaza-1,13-bis[2-(hydroxy)-ethyl]-tridecane (18) as well as 9-benzyl-3,6,12,15-tetrakis(4-nitrobenzenesulfonyl)-3,6,9,12,15-pentaazaheptadecane-1,17-diyldimethanesulfonate (19) were described. The molecular structures of compounds 4-benzyldiethylentriamine-dihydrochloride (3∙2HCl), 4-methyl-N,N-bis(2-(4-methylbenzenesulfonamido)ethyl)benzensulfonamide (26) and 4-benzyl-1,7-bis-(4-toluenesulfonyl)-diethylentriamine (27) were confirmed by X-ray analysis. Via a [2+2]-cyclization of two fragments about the same size (4 and 2,2'-(4-nitro-benzenesulfonamido)bis(ethan-2,1-diyl)dimethansulfonate (16)) compound 20 was also obtained even with lower yield. In the same way the mixed Bn/Ns/Ts protected compound 1,13-dibenzyl-4,10,16,22-tetrakis(4-nitrobenzenesulfonyl)-7,19-ditosyl-1,4,7,10,13,16,19,22-octaazacyclotetracosane (41) and the Bn/Ts protected compound 1,13-dibenzyl-4,7,10,16,19,22-hexatosyl-1,4,7,10,13,16,19,22-octaazacyclotetracosane (38) were prepared. This reaction provides simultaneously the [1+1] cyclization products 1-benzyl-4,7,10-tritosyl-1,4,7,10-tetraazacyclododecane (35), 1-benzyl-4,7,10-tris(4-nitrobenzenesulfonyl)-1,4,7,10-tetraazacyclododecane (39) and 1-benzyl-4,10-bis(4-nitrobenzenesulfonyl)-7-tosyl-1,4,7,10-tetraazacyclododecane (40). The molecular structure of compound 35 was identified. The cyclen ring system exists in the energetically favourable [3,3,3,3]-conformation. The similar constructed compound 40, substituted by heptyl at N10, was determined by X-ray analysis, too. The compound 27 reacts with uranyl nitrate as a potential templat reagens not via cyclization, but redox chemically to N,N-bis(2-(4-methylbenzenesulfonamido)ethyl)formamide (43) with simultaneous building of uran oxid. Compound 43 was also characterized by x-ray analysis. Mass spectroscopy reveals mesomeric and tautomeric forms in the equilibrium. Two mono-N-functionalized three-fold acetic acid alkylated DO3A derivatives, 10-(2-propenyl)-1,4,7,10-tetraazacyclododecane-1,4,7-triacetic acid (48) and 10-(3-butenyl)-1,4,7,10-tetraazacyclododecane-1,4,7-triacetic acid (55) were synthesized and connected with the metal ions of samarium, lutetium and thulium the corresponding complexes [10-(2-propenyl)-1,4,7,10-tetraazacyclododecane-1,4,7-triacetato]samarium (49), [10-(2-propenyl)-1,4,7,10-tetraazacyclododecane-1,4,7-triacetato]lutetium (50), [10-(2-propenyl)-1,4,7,10-tetraazacyclododecane-1,4,7-triacetato]thulium (51) and [10-(3-butenyl)-1,4,7,10-tetraazacyclododecane-1,4,7-triacetato]thulium (56) were gained. Two tetra-N-functionalised cyclen derivatives were built as ligands. The 1,4,7,10-tetra-2-propenyl-1,4,7,10-tetraazacyclododecane (57) was converted to complexes with the metals of nickel (58), copper (61), thulium (62), iron (63) cobalt (64), lithium (60) and rhodium (65). Complex 58 provides after reaction with copper chlo-ride an olefinic π-complex (59). The solid state structure was clarifies as well as the structures of 58 and 61. The ligand 1,4,7,10-tetra(3-butenyl)-1,4,7,10-tetraazacyclododecane (66) provides after conversion with nickel, thu-lium and iron the complexes 67, 68 and 69. The x-ray analysis of 67 elucidates its solid state structure, too. In the particular structures the coordination geometry is a pyramidal surrounding of the metal ions and the distance to the 4N basal plane represents 0.5 to 0.8 Å. A tri-N-functionalised ligand, 1,4,7-tri-(2-pyridyl)-1,4,7,10-tetraazacyclododecane (70) was produced and conversed with thulium chloride to compound 71. Each of the synthesized thulium complexes show fluorescent properties, which were examined by fluorescence spectroscopy and the results were compared. Cyclovoltammograms of the iron-, cobalt- and rhodium- compounds in acteoni-tril were detected in order to analyse the redox chemical properties of these compounds.en
