Das Biosynthesegencluster und Geninaktivierungsstudien des Polyketidantibiotikums atrop-Abyssomicin C

dc.description.abstractIm Rahmen der vorliegenden Arbeit sollte das Biosynthesegencluster des Polyketidanti-biotikums atrop-Abyssomicin C identifiziert, sequenziert und durch Geninaktivierungs-mutagenese untersucht werden. Dies versprach Einblick in die Biosynthese der Tetron-säureantibiotika im Allgemeinen, zu denen die Abyssomicine gehören, sowie in die Biosynthese von atrop-Abyssomicin C im Speziellen. Aus einer Cosmidbank wurden mehrere Cosmide identifiziert, welche Teile des Biosynthesegenclusters für atrop-Abyssomicin C beinhalten. Ein Cosmid wurde vollständig sequenziert (C17), zwei weite-re Cosmide (C45 und C49) wurden mittels Primer-Walking teilweise sequenziert. Das vorliegende Contig (überlappendes DNA-Stück) von 57kb enthält 25 ORFs. Hiervon sind wahrscheinlich 24 für die Biosynthese von Abyssomicin C notwendig. Das Contig um-fasst drei Polyketidsynthase-Gene (abyB1-B3), fünf Gene, welche für die Tetronsäurebi-osynthese notwendig sind (abyA1-A5) und bereits in anderen Biosynthesegenclustern für Tetronsäureantibiotika gefunden wurden, ein Monooxygenase-Gen (abyE), ein drei Gene umfassendes Cytochrom P450 Redox-Set (orfV, W und Z), ein einzelnes Cytochrom P450-Gen (orfX), ein vier Gene umfassendes ABC-Transporter-Set (abyF1-F4), ein weite-res Transporter-Gen (abyD), drei Regulator-Gene (abyC, abyH, abyI), eine Thioesterase Typ II (orfT), eine Zink-bindende Alkoholdehydrogenase (orfU) sowie ein Gen unbe-kannter Funktion (abyK). Die Cluster-Ränder sind noch nicht abschließend untersucht, orfX (am 5’-Ende der Sequenz) und orfU (am 3’-Ende der Sequenz) sind wahrscheinlich nicht mehr Teil des Abyssomicin-Genclusters. Die Gene abyB1–B3 enthalten sieben PKS-Module, fünf codieren für den Einbau von Acetat, zwei für den Einbau von Propionat. Einzelne Deletion der Gene abyI, abyA1, abyA2, abyA3 und abyA4 führte zu einem kom-pletten Ausfall der atrop-Abyssomicin C Biosynthese, was die Notwendigkeit dieser Ge-ne für die Abyssomicin-Biosynthese bestätigt. Die Deletion des Gens abyD führte zu ei-ner Verringerung der Produktion auf ca. 10% des Wildtyp-Niveaus. Es ist sehr wahrscheinlich, dass dieser Transporter nicht alleine die Selbstresistenz von Verruco-sispora AB-18-032 bewirkt. Die Deletion von abyE führte zu einer Reduktion der Abys-somicin-Produktion auf ca. 10% des Wildtyp-Niveaus. Die Biosynthesehypothese für atrop-Abyssomicin C umfasst nach der Bildung eines linearen Vorläufers durch die Poly-ketidsynthase die Biosynthese der Tetronsäure durch AbyA1-A5 und eine anschließende Diels-Alder-Reaktion. Das Diels-Alder-Produkt wird entweder durch AbyE oxidiert, worauf sich ein nukleophiler Angriff durch eine Hydroxygruppe der Tetronsäure an das Epoxid anschließt, was zur Bildung der fertigen Spiroverbindung mit vier Ringen führt. Alternativ könnte OrfV die Hydroxylierung des Diels-Alder-Produkts katalysieren. Im Kulturfiltrat der Mutanten VerrucosisporaabyE und Verrucosispora pK18mob2[orfV] konnten keine Biosyntheseintermediate gefunden werden. In der vorliegenden Arbeit wurde das Biosynthesegencluster für atrop-Abyssomicin C erfolgreich identifiziert, se-quenziert und mittels bioinformatischer Analyse sowie Geninaktivierungs- und Gendele-tionsexperimenten charakterisiert. Darüber hinaus wurde eine Biosynthesehypothese aufgestellt. Weitere Experimente werden nötig sein, um die Biosynthese von atrop-Abyssomicin C im Speziellen und Tetronsäureantibiotika im Allgemeinen abschließend zu klären.de
dc.description.abstractIn the present work, the biosynthetic gene cluster of the polyketide antibiotic atrop- abyssomicin C should be identified, sequenced and investigated by gene inactivation mutagenesis. This promised insight into the biosynthesis of tetronic acid antibiotics in general, which include the abyssomicins, and in the biosynthesis of atrop-abyssomicin C in particular. From a cosmid-bank several cosmids were identified which contained parts of the biosynthetic gene cluster for atrop-abyssomicin C. One cosmid was fully sequenced (C17), two further cosmids (C45 and C49) were partially sequenced via chromosomal walking. The total contig (overlapping DNA pieces) contains 25 ORFs of 57kb. Of these, 24 are probably necessary for the biosynthesis of abyssomicin C. The final contig includes three polyketide synthase genes (abyB1-B3), five genes that are necessary for tetronic acid biosynthesis (abyA1-A5) and were already found in other biosynthetic gene clusters for tetronic acid antibiotics, one monooxygenase gene (abyE), three genes coding for a cytochrome P450 redox set (orfV, W and Z), a single cytochrome P450 gene (orfX), four genes coding for an ABC transporter set (abyF1-F4), one transporter gene (abyD ), three regulatory genes (abyC, abyH, abyI), one type II thioesterase (orfT), a zinc-binding alcohol dehydrogenase (orfU) and a gene with unidentified function (abyK). The cluster boundaries are not yet fully investigated, orfX (5'-end of the sequence) and orfU (at the 3 'end of the sequence) are probably not part of the abyssomicin gene cluster. The genes abyB1-B3 encode seven PKS modules, five for the incorporation of acetate, two for the incorporation of propionate. Individual deletion of genes abyI, abyA1, abyA2, abyA3 and abyA4 led to a complete loss of atrop-abyssomicin C biosynthesis, which indicates the involvement of these genes in abyssomicin biosynthesis. The deletion of the gene abyD led to a drop of production to about 10% of wild-type levels. It is very likely that this transporter is not the only gene conferring self resistance of Verrucosispora AB-18-032. The deletion of abyE led to a reduction of abyssomicin production to about 10% of wild-type levels. The hypothesis for the biosynthesis of atrop-abyssomicin C includes the formation of a linear precursor by the polyketide synthase followed by the formation of the tetronic acid moiety catalyzed by AbyA1-A5 and a subsequent Diels-Alder reaction. The Diels-Alder product could be oxidized by abyE, after which a nucleophilic attack to the epoxide by a hydroxy group could follow, leading to the formation of the final spiro-compound with four rings. Alternatively, OrfV could catalyze the hydroxylation of the Diels-Alder product. In the culture filtrates of VerrucosisporaabyE and Verrucosispora::pK18mob2[orfV], no intermediates of biosynthesis were found. In the present work, the biosynthetic gene cluster for atrop-abyssomicin C was successfully identified, sequenced, and characterized through bioinformatic analysis, genetic inactivation and gene deletion experiments. In addition, a hypothesis for atrop-abyssomicin biosynthesis is presented. More experiments will be needed to resolve conclusively the biosynthesis of atrop-abyssomicin C in particular and tetronic acid antibiotics in general.en
